The study examined whether hydroxychloroquine may be useful in controlling the cytokine storm that occurs late-phase in critically ill SARS-CoV-2 infected patients. The pharmacological activity of chloroquine and hydroxychloroquine was tested using SARS-CoV-2 infected Vero cells and Hydroxychloroquine (EC50=0.72 μM) was found to be more potent than chloroquine (EC50 =5.47=5.47 μM) in vitro. A loading dose of 400 mg twice daily of hydroxychloroquine sulphate given orally, followed by a maintenance dose of 200 mg given twice daily for 4 days is recommended for SARS-CoV2 infection. - Science Translational Medicine – Broad-spectrum antiviral GS-5734 inhibits both epidemic and zoonotic coronaviruses
The study shows that a nucleotide prodrug GS-5734, which is currently in clinical development for the treatment of Ebola, can inhibit SARS-CoV and MERS CoV replication. GS-5734 was also effective against bat-CoVs, prepandemic bat-CoVs and circulating contemporary human CoV in primary human lung cells.
